C5 can be measured in a functional assay using C5-deficient mouse serum as a source of other C components. C5-deficient mice are readily available and the serum can be obtained commercially.

In early atherosclerosis, underlying mechanisms remain incompletely defined, and reliable plasma biomarkers are lacking. In this study, deep quantitative proteomics of human aortic tissues, including healthy segments and regions exhibiting fatty streaks or fibro-lipidic plaques, revealed marked alterations. Proteins involved in lipid transport, complement activation, immunoglobulin functions, and hemostasis were upregulated, especially within the intima of fibro-lipidic lesions. Complement component C5 was confirmed by immunoblot, ELISA, and immunohistochemistry and linked to local activation. Elevated plasma C5 correlated with plaque burden and coronary calcification in two independent cohorts, highlighting its promise as a novel biomarker for subclinical atherosclerosis.
