In a series of purification steps encompassing euglobulin precipitation, DEAE ion-exchange chromatography, and gel filtration, human complement proteins C5 and C3 were isolated with yields of approximately 41% and 20%, respectively. Employing Phenyl–Sepharose chromatography enabled complete resolution, as C3 was associated weakly with the resin while C5 adhered tenaciously, requiring a 50% glycerin solution for elution. Subsequent size exclusion chromatography efficiently eliminated co-purified contaminants, notably C4bp and H, from C5 fractions. The homogeneity of both proteins was validated by SDS–PAGE and immunochemical assessments, revealing that each comprises two disulfide-bridged polypeptide chains. This refined protocol underscores methodology precision and reliability.
