Researchers investigated the roles of complement effectors in collagen antibody-induced arthritis (CAIA) in mice, focusing on C3a and C5a receptors alongside the membrane attack complex. Histopathological and immunohistochemical analyses revealed that clinical severity was reduced by 52% in C3aR–/–, 94% in C5aR–/–, and 65% in C6-deficient mice relative to wild types, with corresponding decreases in tissue injury and IgG/C3 deposition. Additionally, synovial neutrophil counts fell across genotypes, while macrophage numbers declined in C3aR–/– and C5aR–/– mice. Synovial cytokine mRNA (TNF-α and IL-1β) levels were also diminished, suggesting a feedback loop that amplifies joint complement activation and inflammation.
