C4BP Purification Protocol

C4b-binding protein (C4BP) acts as a critical inhibitor of the classical as well as lectin pathways by limiting the effects of activated C4b. It is worth noting that C4BP plays a critical role in limiting the generation of the C3 convertase C4b2a. C4BP is closely linked to the natural decomposition of C3 convertase. These functions allow C4BP to serve as the primary target for pathogens to evade complement attacks. C4BP Purification Protocol

C4BP is a soluble 570 kDa polyplasmic glycoprotein expressed primarily in the liver. It is composed of several identical α-chains and a β-chain, which are mainly composed of the specific domain. The concentration of C4BP in plasma is about 200 mg/L. Here, we describe a simple protocol to purify C4BP from human plasma.
