Published Data
C8 assays focus on MAC-mediated cytotoxicity, employing liposome-based models loaded with fluorescent markers. Upon C8 incorporation into C5b-7 complexes, MAC pore formation triggers dye release, measured via real-time fluorescence quenching. Dose-response curves establish C8 potency (EC50 ≤15 nM in optimized systems). Cross-validation with Western blotting confirms C8 βchain integration into MAC structures. Notably, residual C8 activity in inhibitor screening correlates with >90% hemolytic inhibition at IC50 values <1 µM, underscoring assay sensitivity.
