Encoded by the C8 gene in humans, complement component 8 (C8) is a component of the complement system involved in the complement system. As a part of the membrane attack complex (MAC), studies have shown that genetic defects in C8 can lead to increased susceptibility to Neisserial infections such as meningitis and gonorrhea. In this case, Creative Biolabs has developed the novel C8 purification protocol for our clients to help them do better research.

The purification process demands careful management of complement proteins like C7 and C8 to preserve their structure and function. Scientists can produce large amounts of glycosylated proteins by combining recombinant expression systems with HEK293 cells. Tailored enzyme mixtures in deglycosylation protocols successfully remove glycan groups from proteins without affecting their binding function, since deglycosylated C2 retains its C4b interaction capability. Glycan contributions to molecular weight (e.g., ~16 kDa for C2) are validated via SDS-PAGE and SEC-MALS, ensuring accurate mass determination.
