Detection Protocol of Complement Receptor 2 on Mouse Splenic B Cells

As a receptor for specific cleavage fragments of complement component 3, complement receptor 2 (CR2, CD21) participates in the transport of immune complexes and retention in immune follicles, lowers the stimulation threshold of B cells, and acts as an essential part of complement-driven immune regulation in humoral immunity. CR2 is only expressed in mouse/human B cells and follicular dendritic cells, so the detection of CR2 can be used to identify and characterize B cell populations.

Here, we briefly describe the protocol for detecting CR2 on mouse splenic B cell subsets. It is worth noting that this protocol can also be extended for the detection of CR2 in cells from other sources (peripheral blood, bone marrow, peritoneal cavity, etc.) with simple modifications. A similar procedure can also identify cells with antibodies that recognize human epitopes.
