Hemolytic Assay Protocol for C1 Complex

The multistep cascade of the classical complement pathway (CCP) is initiated and activated by the first component C1 sequence. Binding of C1q to its target results in the stimulation of C1, which in turn activates a downstream multistep cascade in an immunoglobulin-dependent (by binding IgG or IgM) or immunoglobulin-independent manner. C1q does not contain enzymatic activity, but is provided entirely by C1r and C1s. The activativation of the C1 complex will cleave and activate C4, initiating the classical pathway of complement. Here, we briefly summarize the protocol for the hemolytic assay of the C1 complex, which can be used as a screening test and supported by functional analysis data for complement-related studies.
