Complement 4 (C4) is a multifunctional β1-globulin that functions in the classical complement pathway. Cleaved C4 forms two fragments of different sizes, C4a and C4b. C4b interacts with a specific fragment of C2 in a Mg2+-dependent manner, thereby opening the downstream signaling pathway. C4 is essential in immune regulation, virus neutralization, prevention of immune complex deposition, and promotion of phagocytosis. A hemolytic test for C4 can provide screening information for complement function testing and activity results. Here, we briefly introduce the hemolytic assay protocol for C4 in order to provide technical reference for your experiments.
