Introduction of β-Catenin

β-Catenin is a multifunction protein encoded by the CTNNB1 gene in humans. As a protein associated with the cytoplasmic region of E-cadherin, β-catenin has been shown to perform two functions: signal transduction in the Wnt/wg pathway and cell-cell adhesion. Wnt/wg signaling results in β-catenin accumulation and transcriptional activation of specific target genes during development. The cytosolic β-catenin elevates the mutations of Wnt/β-catenin pathway core components in multiple cancer cells, including colon cancer, melanoma, ovarian cancer, hepatocellular carcinoma, endometrial cancer, and prostate cancer.

Clinical Value of β-Catenin in Cancer Therapy

In addition to being considered a potential prognostic biomarker, β-catenin is also an important molecular target in cervical cancer therapy. Besides, nuclear β-catenin expression is related to chemo-resistance and radio-resistance in cervical cancer. There are several approaches for targeting β-catenin that have been developed, including enzyme inhibitors, β-catenin/TCF antagonists, and transcriptional regulators. Some compounds have already progressed to phase II oncology trials, but some are still in the discovery phase. In summary, β-catenin is a potentially druggable target.
