While antibody-secreting cells (ASCs) are a rich source of high-affinity antibodies, current discovery methods are often limited by low throughput, high cost, or technical complexity, making them inaccessible. This publication introduces a straightforward, high-throughput technology for the rapid discovery of monoclonal antibodies directly from ASCs. The method integrates microfluidic encapsulation of single cells into an antibody capture hydrogel with antigen bait sorting via conventional flow cytometry. Utilizing this approach, researchers screened millions of mouse and human ASCs, successfully obtaining high-affinity and potent neutralizing monoclonal antibodies against SARS-CoV-2 within two weeks, achieving a high hit rate. This technology streamlines antibody discovery and immunological studies by enabling efficient access to the underexplored ASC compartment.
