For CFH functional analysis, several activities can be measured. For example, the decay-acceleration activity refers to the ability to displace Bb from C3b to inactivate the C3 convertase C3bBb, Factor I-cofactor activity refers to the ability to form complexes with C3b, while classical pathway downregulation refers to the ability to compete with C1 or C1q for binding targets such as cardiolipin. Here, Creative Biolabs has developed the CFH functional analysis protocol for our clients all over the world.

CFH dysfunction is implicated in atypical hemolytic uremic syndrome (aHUS) and age-related macular degeneration. Functional assays now incorporate patient-derived variants to map structural domains critical for C3b binding or decay-accelerating activity. For example, mutations in SCR domains 19–20 disrupt CFH-cell surface interactions, a finding validated by reduced binding in flow cytometry assays. Such data inform therapeutic strategies, including monoclonal antibodies or recombinant CFH supplements.
