Complement Receptor 1 (CR1/CD35) Purification Protocol

Complement receptor type 1 (CR1), also known as CD35, is a 210-290 kDa membrane-bound glycoprotein with specificity for C3b and C4b. CR1 is a key factor in the activation of the plasma protein complement system. CR1 shares structural similarities with various proteins, such as factor H, decay accelerating factor (DAF), IL-2 receptor and β2-glycoprotein 1, many of which act as key regulators of complement activity. CR1 is a multifunctional protein that is present in different cells, such as monocytes, macrophages and dendritic cells. On neutrophils and monocytes, it is intimately associated with the adhesion of C4b/C3b-binding ligands. CR1 on B lymphocytes plays a crucial role in promoting antigen presentation to T cells. In addition, CR1 is involved in various diseases. In recent years, CR1 has gradually emerged as an attractive molecule for gaining insight into the pathology, diagnosis and treatment of many diseases. CR1 Purification Protocol

Although erythrocytes have a low level of CR1 expression, they remind a good choice to obtain CR1 due to their availability and relatively easy preparation of membrane extracts. Here we describe a simple procedure for the purification of CR1 from erythrocytes.
