Introduction | Complement component 5a (C5a) is an inflammatory polypeptide produced from cleavage of complement component C5 by protease C5-convertase. Biologically, C5a is a potent anaphylatoxin and a chemotactic factor playing a critical role in inflammation. It has been demonstrated that C5a could increase vascular permeability and induce chemotaxis and the release of active mediators. In human plasma or serum, C5a is rapidly "inactivated" or converted to C5a desArg by serum carboxypeptidase N through the removal of the C-terminal arginine (Arg) within minutes. Although C5a des-Arg is considered to be the inactivated form of C5a, C5a desArg retains significant biological activity including weak anaphylatoxic and chemotactic activities, inducing muscle contraction and vascular permeability increase, causing the release of histamine and lysosomal degranulation of immune cells. As C5a desArg is the main active form of C5a in plasma and serum, the quantitation of C5a-desArg in samples is an indirect reflection or determination of the level of complement activation in the test samples. |
