MBL Purification Protocol

Mannose-binding lectin (MBL) is a serine protease that plays a crucial role in innate immunity as an opsonin. With unique recognition capabilities, MBL can bind to the surface of a variety of pathogenic microorganisms, including bacteria, viruses, protozoa and fungi, leading to the activation of the lectin pathway of the complement system. Another crucial function of MBL is that the molecule binds to senescent and apoptotic cells and enhances the phagocytosis of apoptotic cells and cellular debris by phagocytic cells. The purification of MBL depends on carbohydrate affinity chromatography and the sequential use of different sugars to remove contaminating anti-carbohydrate antibodies from MBL preparations.
