The cDNA of hamster complement C1s was subcloned into the expression vector BCMGSNeo and introduced into SEA7 cells, a polyoma virus-transformed A31 mouse fibroblast line. The transfected cells secreted significant amounts of recombinant C1s, which became activated in a serum-free medium and exhibited hydrolytic activity on acetyl-Gly-l-Lys-naphthyl ester (AGLNE). Purification of C1s to homogeneity from the culture medium involved size-exclusion HPLC and other methods. The purified hamster C1s was capable of depleting human complement in hemolytic assays and hydrolyzing gelatin as demonstrated in enzymographic studies.
