Complement C5a has been identified as a key player in tissue regeneration, acting via its receptor C5aR, which is expressed in the nervous system—a region where therapeutic options for repair remain limited. Researchers investigated the influence of C5a on axonal regrowth using an in vitro axotomy paradigm. Primary hippocampal neurons from embryonic rats were isolated; RT-PCR confirmed C5aR mRNA, while immunofluorescence demonstrated membrane localization, NF-L co-localization in mature neurons, and receptor phosphorylation triggered by C5a. Administering C5a in a 3D-printed microfluidic device significantly accelerated both axonal growth speed and extent, suggesting a promising regenerative role with therapeutic potential in central nervous system injuries.
