In leukocytes, C3a and C5a trigger chemotaxis through a Gi-dependent mechanism that is susceptible to inhibition by pertussis toxin. Using a fluid-phase convertase system comprising cobra venom factor (CVF) and Bb, human C3a and C5a were derived from C3 and C5 and subsequently purified via DEAE chromatography with an imidazole HCl/NaCl buffer, followed by adsorption and NaCl elution. Remarkably, only C5a induced chemotactic migration in immortalized human dermal microvascular endothelial cells. This response was impeded by PI3-kinase, src kinase, and EGFR inhibitors, as well as neutralizing antibodies against EGFR and heparin-binding EGF-like factor, with C5a stimulation promoting EGFR phosphorylation.
