Introduction of RAGE

Receptor for advanced glycation end-product (RAGE), also called AGER, is a 35 kDa multiligand transmembrane receptor of the immunoglobulin gene superfamily. Full-length RAGE contains a V-type domain, which functions for ligand binding, two C-type domains, a transmembrane spinning helix, and a C-terminal cytosolic domain (ctRAGE) which is required for signal transduction. In addition, the receptor for RAGE is associated with a variety of signaling molecules such as advanced glycation end products (AGEs), HMGB1, phosphatidylserine, C3a and advanced oxidation protein products (AOPPs). Generally, it is widely localized in many cell types such as smooth muscle cells, hepatocytes, neurons, endothelial cells, and monocytes. Importantly, RAGE regulates a number of cell processes of pivotal importance like inflammation, apoptosis, proliferation and autophagy.

RAGE Marker of Sepsis

Sepsis is a complex clinical syndrome resulting from a harmful host inflammatory response to infection. However, it is still a major cause of morbidity and mortality in adult and pediatric intensive care units. Studies have shown that bacterial components, such as lipopolysaccharide (LPS), could lead to the activation of an inflammatory cascade which can result in the release of inflammatory mediators during the development of sepsis. The overproduction of inflammatory mediators can lead to endothelial and epithelial injury, vascular leakage, edema, and vasodilatation. It has been reported that RAGE is usually abnormally up-regulated and plays crucial roles in the development of a number of human illnesses, such as diabetes, cardiovascular diseases, and ischemic injury. Most importantly, the serum level of sRAGE (soluble isoforms of RAGE) is significantly elevated with the severity of sepsis. In this case, RAGE can be used as a useful marker for the detection of sepsis.
