RANTES

RANTES, also known as activation-regulated normal T cell expression and secretion factors, is a member of the CC cell subfamily of chemotaxis cells. RANTES is a small molecule protein with a molecular weight of 8 kD. There is no glycosylation site at the N-terminus. Its three-dimensional monomer structure is highly conserved. Magnetic resonance spectroscopy shows that the monomer of RANTES contains a C-terminal helix, three-stranded antiparallel sheet layers, and two N-terminal short stocks. It has a wide range of sources, most tissues can release RANTES under the stimulation of the inflammatory factor IL-1β or TNF-α, and RANTES is secreted by CD8+ T cells, epithelial cells, fibroblasts, and platelets. Due to its ability to induce infiltration of leukocytes into sites of inflammation, RANTES was initially considered to be a typical chemokine cytokine. Besides, RANTES is a potent leukocyte activator involved in many inflammatory reactions, and it is highly expressed in various tumors including hepatocellular carcinoma. RANTES not only inhibits the replication of the HIV virus but also enhances replication under certain circumstances.

RANTES as Marker of Sepsis

HMGB1 is described as a central mediator of host response to sepsis, platelets play an important role in the host's response to bacterial infections, and platelet HMGB1 is essential for effective bacterial clearance in the intra-abdominal polymicrobial sepsis model. Studies have shown that HMGB1 is essential for platelet activation and also regulates neutrophil chemotaxis and ROS production by modulating platelet activation. Platelet HMGB1 regulates neutrophil recruitment and platelet-neutrophil interactions by releasing the platelet-derived chemokine RANTES. Among them, RANTES is a component of platelet alpha particles and is essential for the recruitment of neutrophils. When platelets are activated, the chemokine RANTES is released to recruit granulocytes to eliminate sepsis bacteria.
