Introduction of NF-κB

Nuclear factor κB (NF-κB) is a ubiquitously expressed transcription factor that belongs to the NF-κB protein family which shares a highly conserved DNA-binding and dimerization domain termed Rel homology region (RHR) that enables them to homo- or heterodimerize. Studies have suggested that NF-κB is able to mediate signal-induced expression of numerous genes involved in different biological processes, including immune responses, inflammation, cell growth, and survival. Generally, NF-κB transcription factors can be activated as a response to a variety of signals, including cytokines, pathogens, injuries, and other stressful conditions. And inappropriate activation of the NF-κB signaling pathways has been linked to autoimmunity, chronic inflammation, and various cancers.

NF-κB Marker of Sepsis

Severe sepsis is a complex syndrome characterized by an amplified host inflammatory response leading to organ dysfunction. During severe infections, NF-κB is crucial for the regulation of immune/inflammatory responses and the control of cell proliferation/apoptosis. It has been reported that inhibition of NF-κB activation results in systemic hypotension, ameliorates septic myocardial dysfunction and vascular derangement, inhibits multiple proinflammatory gene expression, and prevents microvascular endothelial leakage. Most importantly, NF-κB activity was significantly higher in human subjects with sepsis. What's more, greater levels of NF-κB activity are associated with a higher rate of mortality and worse clinical outcome. In this case, the level of NF-κB can be used as a specific marker for the diagnosis of sepsis.
