Introduction of Toll-like Receptor 2

Toll-like receptor 2, also known as TLR2, is a protein encoded by the human TLR2 gene that belongs to the Toll-like receptor (TLR) family. As a membrane surface receptor, TLR2 plays a fundamental role in pathogen recognition and activation of innate immunity. It can recognize pathogen-associated molecular patterns (PAMPs) such as bacterial, fungal, viral, and certain endogenous substances that are expressed on infectious agents. In addition, TLR2 has the ability to mediate the production of cytokines necessary for the development of effective immunity. Generally, TLR2 is found in the epithelia of air passages, pulmonary alveoli, renal tubules, and the Bowman's capsules in renal corpuscles. Moreover, TLR2 is also expressed by intestinal epithelial cells and subsets of lamina propria mononuclear cells in the gastrointestinal tract.

Toll-like Receptor 2 Marker of Sepsis

As a member of immune receptor family, TLR2 triggers pro-inflammatory responses following recognition of a range of microorganisms such as bacteria, viruses, and fungi. It is mainly expressed by myeloid cells and capable of down-modulating TLR-mediated pro-inflammatory responses in response to microbial challenges. It has been reported that plasma level of TLR2 is elevated in a number of infective and inflammatory conditions as it constitutes an important first-line negative regulatory mechanism to avoid harmful inflammatory responses. Most importantly, studies have suggested that the activation of TLRs, especially TLR2, is responsible for the inflammatory responses in sepsis and the serum level of TLR2 is significantly higher than that of healthy people. Under this condition, TLR2 can be used as a specific marker for the detection of sepsis.
