Importance of Glycosylation

Glycosylation of proteins is one of the most important post-translational modifications (PTMs), with more than 50% of all human proteins estimated to be glycosylated. The site-specific nature of this modification is involved in numerous essential biological functions including cell proliferation, differentiation and migration, cell-cell recognition and interaction, and signal processes that regulate human development, immunity and disease. The glycans found in humans are predominantly attached to the protein in two ways: 1) N-glycan, a GlcNAc residue is added to Asn within Asn-X-Ser/Thr, where X can be any amino acid except proline; 2) O-glycan, a GalNAc residue is attached to the hydroxyl group of Ser or Thr. Glycoproteins on the cell surface typically undergo glycosylation in the endoplasmic reticulum-Golgi pathway where a sequence of regulated events occurs, resulting in creating highly complex but specific glycan structures on the proteins.
