Glycoproteins Analysis Based on RP-HPLC

RP-HPLC has a non-polar stationary phase and an aqueous, moderately polar mobile phase. This method is very efficient to separate the polar compounds due to the interaction between polar compounds with polar mobile phase. RP-HPLC experimental system is usually composed of an n-alkylsilica-based sorbent. Solutes are eluted from sorbent with gradients of increasing concentrations of an organic solvent such as acetonitrile containing an anionic modifier. Complex mixtures of peptides and proteins can be routinely separated. Separations can be easily performed by changing the gradient slope of solvents, the operating temperature, the ionic modifier, or the organic solvent composition. The separated fractions can be used for further characterization.

RP-HPLC is a commonly used method for the analysis and purification of peptides, proteins, and glycoproteins. Monosaccharide composition and content can be determined by using the RP-HPLC separation of p-aminobenzoic ethyl ester derivatives of neutral and amino sugars released from glycoproteins. Moreover, a study has shown that RP-HPLC method has the best coverage for glycopeptide analysis based on the number of detected glycosylation sites and has identified glycopeptides in three model glycoproteins, superior to chromatographic and/or enrichment approaches such as HILIC, and lectin affinity.
