Introduction to Internalization

Generally speaking, there are 2 kinds of internalization formats: receptor-mediated and non-receptor-mediated. The former one is the most conventional internalization pattern. Upon binding with membrane receptors, the target molecule can translocate into the cytosol in endosomes, and subsequently undergoes a complex array of recycling / degradative pathways. This procedure is considered to correlate with many factors, such as target receptor, binding affinity, etc.

Internalizing antibodies have been shown to promote or inhibit cell growth, mainly relying on the epitope recognized. Inhibitory compounds can be used as a cure for cancer or for other conditions characterized by cell hyperproliferation, and for treating inflammation (anti-inflammatories). Stimulatory antibodies could be used for stimulating the growth of relevant cells like stem cells.

Magic™ Whole Fluorescent Stable Cell Lines

We now offer a comprehensive set of stable transfected cell lines for high-throughput screening of internalizing compounds. These cell lines stably express high levels of target receptors (e.g. GPCRs, ion channels) fused with a fluorescent biosensor protein. These cell lines are designed to assay compounds or analyze stimuli for their ability to modulate receptor internalization by tracking cellular fluorescence intensity & location, which can make excellent tools for internalizing antibodies screening and validation. There are more than 20 ready-to-use stable cell lines available in Creative Biolabs for internalization assessment (as shown below):
