Introduction of VLP Production in E. coli Cell Expression System

The most popular bacterial host cell for VLP production is Escherichia coli. An expression system based on E. coli has many benefits, such as low cost of production, fast cell growth, high protein expression level, and easy scaling-up. However, because of a number of issues, including incomplete disulfide bond formation, lack of PTM system, and issues with protein solubility, E. coli platforms are not suitable for producing enveloped VLPs. On the other hand, E. coli expression systems are appropriate for producing non-enveloped VLPs with one or two viral structural proteins. Many VLP vaccines against infectious and non-infectious disorders have entered clinical trials. These vaccines were produced using E. coli expression systems. Apart from E. coli, certain additional bacterial species have also been shown to successfully generate VLP.
