Case Study: Discovery of pH-Dependent scFv via Premade Phage Display Library

Objective To identify antibody fragments (scFvs) with pH-dependent binding properties using our premade human scFv library.
Approach Three consecutive rounds of phage display selection were performed using the target antigen.
Elution strategy: Low-pH buffer was used to selectively elute pH-sensitive binders.
Control: Trypsin-based elution served as a benchmark for comparison.
Screening & Expression Preliminary screening identified several pH-responsive scFv candidates.
These were recombinantly expressed in small scale for functional validation.
