Background

Cytotoxic T lymphocytes (CTL) are important effector cells of the immune system. By killing target cells on the basis of cell-surface antigen recognition, they play important roles in the host immune response to tumors, transplants, and viruses. Studies have shown that T cells perform their functions by elaborating a number of distinct effector mechanisms. What’s more, in vitro cytotoxicity assays have shown the presence of two major contact-dependent cytotoxic pathways. First, cytotoxic effect cells can secrete lytic granules, comprising a pore-forming toxin, perforin, and pro-apoptotic serine proteases, granzymes, which synergistically kill target cells by activating various lytic pathways. Second, effector cells produce the TNF family members, such as TNF-α, Fas ligand (FasL) or TRAIL, thereby inducing multimerization of their cognate receptors on target cells and leading to apoptosis.

T Cell Cytotoxicity Assay Provided by Creative Biolabs

Traditionally, scientists use MTT, CCK8, and CFSE-PI to detect cytotoxicity. However, as T cells also undergo apoptosis when incubated with tumor cells, these methods are unable to detect T cell cytotoxicity.
