Introduction to TIM3

T-cell immunoglobulin and mucin-domain-containing-3 (TIM-3), also known as hepatitis A cellular receptor 2, belongs to the TIM family cell surface receptor proteins that share a similar structure, in which the extracellular region consists of membrane distal single variable immunoglobulin domain (IgV) and a glycosylated mucin domain of variable length located closer to the membrane. This protein is an immune checkpoint expressed by subtypes of lymphoid cells, such as lymphocytes Th1, Th17, Tc1, NK, as well as by myeloid cells. TIM-3 has reported being involved in the mediation of CD8+ T-cell exhaustion together with other inhibitory receptors including PD-1 and LAG-3. Furthermore, this protein is also a Th1-specific cell surface protein which regulates macrophage activation, suppresses Th1-mediated auto- and alloimmune responses, and enhances immunological tolerance. Studies have reported TIM-3 regulates immune tolerance in mouse models of infectious diseases, alloimmunity, autoimmunity, and tumor immunity. Therefore, TIM-3 seems to be a promising target for current immunotherapy.

Background: Multiple myeloma (MM) is a cancer that originates in the plasma cells within the bone marrow. Although treatment options have improved, it continues to be a difficult disease to fully eradicate. One promising approach in cancer therapy is harnessing the power of natural killer (NK) cells, which have shown strong anti-tumor activity and potential in immunotherapy for various cancers, including MM.

Purpose: To investigate the impact of inhibiting the Tim-3 pathway on the ability of NK cells to target multiple myeloma (MM) in laboratory settings and live subjects, researchers utilized antibodies targeting Tim-3 and its ligands in this study.

Results: The findings indicate that targeting Tim-3 could be a promising approach for developing antibody- or NK cell-based immunotherapies against multiple myeloma.
