Cytotoxicity Assay

TCR is a molecule expressed on T lymphocyte surface that mediates antigen-specific recognition and reactivity through presenting antigenic peptides to the major histocompatibility complex (MHC). Introducing genes encoding a tumor-reactive TCR could be used to generate T cells toward an antigen of interest, thereby establishing tumor antigen-specific T cells which are used to infuse to the patient. Transgenic TCRs are capable of recognizing both extracellular and intracellular antigens that need to be presented, which means it can target a large scale of tumor antigens. Cytotoxicity refers to the ability of certain chemicals or mediator cells to destroy living cells. By using a cytotoxic compound, healthy living cells can be induced to undergo necrosis. Given this information, a successful engineered T cell with antigen-specific TCR can harness their own cytotoxicity to the target and eradicate tumors or other disease cells. Therefore, an accurate measurement of cytotoxicity is proven to be a very valuable index in TCRs in vitro assay that might pose certain health risks in patients.

Creative Biolabs has developed various assay methods available for customers to measure the cytotoxicity of TCR-modified T cells. We always introduce vital dyes, protease biomarkers or by measuring ATP content to our clients. The formazan dyes are chromogenic products formed by the reduction of tetrazolium salts by dehydrogenases, which are released at cell death. We also monitor cytotoxicity by the use of the SRB and WST-1 assay which are high through methods.
