TCR is a molecule expressed on T lymphocyte surface that mediates antigen-specific recognition and reactivity through presenting antigenic peptides to the MHC. TCR heterodimers consist of α and β chains or γ and δ chains. Introducing genes encoding a tumor-reactive TCR could be used to generate T cells toward an antigen of interest, thereby establishing tumor antigen-specific T cells which are used to infuse to the patient. In order to alter T-cell specificity, genetically modified TCR therapies are developed. They are achieved by the expression of specific TCR α and β chains to mediate the antigen-recognition process. One powerful approach to examine T-cell receptor (TCR) properties and T-cell function is to clone TCR genes into retroviral vectors and use them to transfer antigen specificity into other cells. One drawback of this approach is that the cloning of the TCR genes into the retroviral vector is a somewhat complicated, time-consuming process.
