T lymphocytes, also known as T cells, recognize foreign antigen through the T cell receptors (TCRs). There are two types of TCRs that has been identified on the surface of T cells, the alpha/beta-TCR and the gamma/delta.-TCR, and the former one is expressed on the majority of T cells that recognize foreign antigens complexed with major histocotnpatibility complex (MHC) molecules on the surface of antigen-presenting cells. Throughout T-cell development and proliferation, the progeny of a single T cell will express the identical TCR and contain the identical TCR gene rearrangement that serves as a molecular marker of clonality, which is a complex and highly regulated process and any dysregulation may lead to disease, such as cancers, immunodeficiency and autoimmunity diseases. Therefore, clonality assay via assessment of TCR gene rearrangements has become a critical and valuable tool in the diagnostic of lympho-proliferations and T cell related diseases.
