Chimeric antigen receptor (CAR) engineered natural killer (NK) cells show great promise against hematological malignancies in recent advances. Multiple methods for genetic engineering of PBMC-derived NK cells and continuously expanding NK-92 cells are well established during the past decade, which facilitates the generation and expansion of novel NK cells with optimized therapeutic efficacy. To enhance the therapeutic utility of CAR-NK cells, Creative Biolabs provides a variety of genetic strategies for engineering next-generation NK cells.

Overview of our NK cell engineering strategies

To overcome some of the limitations of NK cell therapy, we have adopted a variety of genetic engineering strategies to address the above mentioned limitations. Specifically, these strategies include activation of exogenous receptor expression or selective down-regulation of inhibitory receptors to improve CAR-NK cytotoxicity; down-regulation of inhibitory cytokines and introduction of dominant negative receptors to reduce tumor environmental sensitivity; expression of chemokine receptors to increase in vivo proliferation and persistence through autocrine cytokine stimulation and tumor homing.
