In the past few years, antibodies have shown promising results in treating various diseases, especially for many types of cancers, in a variety of preclinical trials or clinical trials. Among them, the agonistic antibody has been considered as a powerful tool for enhancing immune responses against infections or diseases. Meanwhile, pilot studies have demonstrated that agonistic monoclonal antibodies can trigger immune responses against tumor cells by activating the function of cells, including T cells, NK cells. In general, the signal activation is related to specific antigens, such as cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) and programmed death 1 (PD-1), as well as many costimulatory molecules, including CD137, OX40, and GITR. Furthermore, researchers have also revealed that interleukin-2 (IL-2) is a key cytokine in mediating lymphocytes, which has been approved for combination therapy in carcinoma and metastatic melanoma. For example, the anti-IL-2 agonistic antibody has been used for boosting anti-tumor effect in CTLA-4 blockade, and many large scales of phase I clinical trials have been conducted to assess the safety of CTLA-4 blockade in combination with an anti-IL-2 agonistic antibody.

As the main member of the cytokine family, IL-2 has been one of the most attractive therapeutic agents for harnessing the immune system to treat malignant tumors. Moreover, a more recent study suggests that IL-2 can regulate the expression of some checkpoint molecules, including PD-1 and CTLA-4, by stimulating T regulatory cells. As a consequence, a wide collection of anti-IL-2 agonistic antibodies basing on checkpoint proteins have been developed and evaluated in a full range of disease treatment.
