Cell adhesion molecule 1 (CADM1, also known as TSLC1) is a single-pass transmembrane adhesion protein of the immunoglobulin superfamily encoded by the CADM1 gene. It is expressed in epithelial, neural, and Sertoli cells and contributes to intercellular junction organization. Unlike classical adhesion molecules such as E-cadherin, CADM1 mediates both homophilic and heterophilic cell-cell adhesion through its extracellular immunoglobulin-like domains and interacts with intracellular scaffold proteins through its cytoplasmic tail. CADM1 contributes to epithelial integrity and regulation of cell migration, and reduced CADM1 expression, including through promoter hypermethylation, has been associated with tumor progression in several cancer types. CADM1 also participates in neural development and male reproductive function, making it an important research target in IgSF-mediated adhesion and tumor biology.
CADM1 mediates intercellular adhesion through extracellular Ig-like domains and links cell-cell contacts to intracellular scaffold and polarity networks. Its adhesion and signaling functions contribute to epithelial homeostasis, neural development, and tissue organization. Loss or reduced expression of CADM1 has been associated with impaired cell adhesion and enhanced malignant phenotypes in multiple tumor models, although the underlying mechanisms are context dependent. Therefore, CADM1 represents an important research target for IgSF adhesion biology and tumor suppressor-associated mechanisms.
