Background

C-C motif chemokine ligand 18 (CCL18) is a soluble secreted chemokine encoded by CCL18 gene, which is mainly secreted by myeloid cells and released into extracellular tissue microenvironment. Different from the integrated transmembrane receptor, CCL18 is a soluble chemotactic ligand without lipid-anchored transmembrane segment and intracellular signal domain. The molecule can interact with chemoreceptor complex on cell surface under physiological steady-state conditions. When the buffer regulatory factors of soluble chemokines are insufficient, it is easy to appear leukocyte recruitment disorder; CCL18 can mediate moderate chemotactic signal buffering effect and maintain the dynamic balance of immune cell migration in inflammatory tissue and steady-state tissue microenvironment. Different tissue regions will produce chemokine combinations with different components, which requires a variety of soluble chemokine pools to ensure the overall distribution of white blood cells in a multicellular tissue system in a balanced state. The free diffusion of CCL18 can continue to bind to the surface receptors of target cells, inhibit abnormal immune cell recruitment behavior, and maintain local tissue immune homeostasis.

The variation of CCL18 gene sequence may change its receptor binding affinity, which is related to the change of leukocyte composition in tissue. In the CC chemokine family, no other molecule can completely duplicate the dual functions of CCL18, which simultaneously binds to receptors and regulates the chemotaxis of myeloid cells. The change of CCl-18 expression level often echoes the immune activation of local tissues, which makes it an important research object to study soluble CC chemokines and leukocyte migration regulation. CCL18 is located in extracellular aqueous solution environment and is not embedded in lipid bilayer; The molecule binds to the receptor complex on the target cell surface, but it does not initiate the transmembrane signal cascade reaction itself. As a fully secreted molecule, CCL18 is different from the membrane-bound effector molecule, and has dual functions of shaping local chemotactic concentration gradient and mediating paracrine immune cell communication. The decline of CCL18 function can disturb the migration pattern of leukocytes and weaken the signal buffering ability mediated by local chemokines, which further confirms its research value in the basic research of soluble chemokines.

CCL18 Protein Function: Core Roles in Receptor Interaction and Chemotactic Signal Coordination

The biological functions of soluble CCL18 chemokine protein are focused on sustained cell-surface receptor interaction and paracrine chemotactic signal coordination:

  • Broad Receptor Complex Affinity: Might interact with multiple leukocyte-originated receptor assemblies without triggering consistent intracellular signal cascades. The chemokine binds receptor components originating from immune-cell surfaces and expands the scope of leukocyte-trafficking regulation within tissue microenvironments.
  • Tissue Immune Regulation: Could moderate excessive chemotactic stimulus outputs to ease local immune-recruitment overload. This regulatory mode prevents drastic leukocyte-migration fluctuation that disrupt stable tissue physiological conditions.
  • Chemotaxis Mediator: Appears to facilitate reversible molecular attachment to cell-surface chemokine receptor complexes. Weak non-covalent ligand-receptor binding generates transient interaction patterns detectable via standard laboratory analytical workflows.
  • Chemokine Gradient Modulation: Shapes local chemokine concentration gradients to coordinate overall leukocyte-recruitment intensities.
  • Research Model Relevance: Sequence variants of CCL18 may shift receptor-binding efficiency within laboratory research systems.

CCL18 Protein Product

Creative Biolabs offers purified CCL18 protein samples produced under unified preparation workflows, including full-length CCL18 constructs and isolated receptor-binding domain variants. Truncated domain fragments cannot support complete receptor-interaction behaviors, while full-length constructs suit research focused on chemokine-receptor association and paracrine chemotaxis observation. All batches receive uniform quality screening. Functional relevant observation may only be carried out with full-length samples under simulated extracellular microenvironment setups. Consistent structural features are preserved across batches to support comparative laboratory analysis across separate test groups. Full-length CCL18 samples retain intact receptor-binding motif conformation after standardized purification, which supports reliable detection of weak and transient ligand-receptor contacts for comparative functional analysis.
