Background

Cadherin 6 (also known as K-cadherin) is a highly conserved type II cadherin encoded by 6(CDH6 gene, which belongs to the transmembrane adhesion core protein dependent on calcium ions, and is widely involved in vertebrate adhesion connection assembly and orderly construction of tissue morphology. CDH6 is specifically expressed in embryonic renal epithelium and various glandular epithelial tissues, and the extracellular domain contains tandem calcium ion binding repeat domains, which can mediate the specific recognition of homologous cells and stabilize intercellular junction. The protein can synergistically regulate cell adhesion strength, intracellular cytoskeleton rearrangement and downstream signal cascade reaction, and ensure the regularity of tissue structure in the process of embryonic organogenesis and postnatal tissue homeostasis maintenance. Compared with other cadherin subtypes with different tissue distribution patterns, CDH6 has unique functional advantages in the regulation of epithelial tissue spatial patterns. Under pathological conditions, abnormal expression level of CDH6 can reshape cell motility and invasion potential, and participate in the abnormal progress of many diseases.

CDH6 relies on calcium-mediated extracellular homologous binding to stabilize the intercellular junction structure, activate intracellular adhesion signal pathway synchronously, coordinate and regulate the collective migration behavior of cells, and maintain the integrity and stability of tissue structure. Its highly conserved extracellular structural motifs can accurately identify homologous cells, continuously start intracellular signal transduction, and maintain the physiological stability of epithelial tissues. CDH6 is widely involved in many basic biological processes, such as embryonic organ development, epithelial cell adhesion construction and long-term tissue homeostasis maintenance. When the expression of CDH6 is unbalanced or its function is damaged, the integrity of intercellular junction structure is destroyed and the cell invasion ability is significantly improved, which is closely related to the malignant progress of many epithelial tumors.
