CEA cell adhesion molecule 6 (CEACAM6) is a glycosylated cell-surface immunoglobulin superfamily protein encoded by the CEACAM6 gene, enriched in epithelial and transformed tumor cell populations, anchored to plasma membrane lipid bilayers. Distinct from homeostatic CEACAM paralogs, CEACAM6 mediates homotypic intercellular crosslinking via conserved extracellular Ig domains while modulating inhibitory immune receptor engagement to dampen anti-tumor effector responses. Under normal epithelial steady states, low CEACAM expression maintains mild intercellular adhesion without immune suppression. Upon malignant transformation, elevated CEACAM6 surface abundance reinforces tumor cell clustering and blocks myeloid-mediated anti-tumor surveillance signals. Unlike constitutive epithelial adhesion factors, CEACAM6 carries tumor-specific migratory and immune-inhibitory functions that cannot be fully compensated by other CEACAM members, linking homotypic cell binding to neoplastic dissemination pathways. Loss of CEACAM6 attenuates tumor invasive capacity, while sustained overexpression accelerates metastasis and immune evasion, marking CEACAM6 as a core research target for tumor adhesion protein and anti-tumor compound screening.
CEACAM6 executes homotypic adhesion and immune modulatory function embedded in cell surface bilayers, utilizing conserved Ig-like extracellular pockets to mediate self-association and interaction with myeloid inhibitory receptors. Conserved binding motifs separate its ligand spectrum from tissue-restricted CEACAM isoforms. CEACAM6-mediated molecular bridging connects tumor cell clustering to suppressive immune signal transduction, balancing invasive potential according to malignant transformation grade. CEACAM6 participates in epithelial tissue adhesion, tumor metastatic spread and local immune suppression. Deficient CEACAM6 expression disrupts neoplastic cell aggregation and weakens immune evasion capacity. Therefore, CEACAM6 represents a pivotal research target for Ig adhesion protein and anti-metastatic compound screening.
