Crumbs cell polar complex component 1(CRB1) is a highly conserved apical single transmembrane scaffold protein encoded by CRB1 gene, which is mainly located in the apical cell membranes of retinal photoreceptor cells and Müller glial cells, and only slightly expressed in the central nervous system. Compared with homologous protein CRB2 and short domain subtype CRB3, CRB1 has a large extracellular domain rich in EGF and laminin globular motifs, and has an intracellular tail structure with PDZ and FERM binding sites, which is the core assembly element of retinal Crumbs polar complex and can stabilize the intercellular junction structure of retinal outer membrane. Under the conditions of retinal development and physiological steady state, CRB1 is continuously and stably expressed, maintaining the standard top-base polar arrangement of photoreceptor cells, stabilizing the intercellular adhesion structure, and effectively avoiding the disorder of retinal layered structure. When the mutation of double allele loss occurs in CRB1, the polar complex assembly system mediated by CRB1 collapses, resulting in disorder of photoreceptor cells and progressive degeneration of retinal pigment, and inducing severe visual impairment diseases such as Lieber's congenital amaurosis and retinitis pigmentosa. Other CRB homologous proteins can't compensate CRB1' unique retinal polarity maintenance function, and CRB1 deletion will completely disrupt the morphogenesis of photoreceptor cells, while the pathogenic mutant CRB1 can't assemble functional polar complexes, making it the core irreplaceable target of retinal epithelial polarity biology and drug screening for hereditary blindness diseases.
CRB1 is anchored on the lipid bilayer of photoreceptor cells and glial cell membranes, and plays the role of top polar scaffold regulation. It mediates homologous interaction of CRB1 and heterologous interaction of CRB1-CRB2 through multi-domain extracellular functional regions. At the same time, it uses intracellular functional motifs to recruit PDZ polar adaptor proteins to assemble stable intercellular junction devices. Its unique extra-long extracellular structure forms obvious functional differentiation with CRB3 lacking ligand binding region, which endows it with exclusive retinal tissue protection function. The polar complex assembly pathway mediated by CRB1 connects the differentiation of embryonic retinal progenitor cells with the structural stability of adult photoreceptor cells, which can dynamically maintain the integrity of retinal layered structure according to the development process and lighting environment. The protein is involved in the maturation and differentiation of photoreceptor cells, the construction of retinal outer membrane and the maintenance of retinal tissue structure for life. The defect of CRB1 expression or function will completely destroy the complete structure of retinal epithelium and trigger the progressive apoptosis of photoreceptor cells. To sum up, CRB1 is the key target for studying the mechanism of ocular polar proteins and developing drugs for hereditary retinopathy.
