Background

CXC motif chemokine ligand 3 (CXCL3) is a secreted inflammatory CXC chemokine encoded by the CXCL3 gene, which can be rapidly induced in epithelial cells, macrophages, and tumor-associated stromal cells under inflammatory and other microenvironmental stimuli. Like the related GRO family chemokines CXCL1 and CXCL2, CXCL3 activates CXCR2 and promotes neutrophil recruitment to inflammatory and tumor-associated tissues. In acute inflammatory responses, transient CXCL3 production contributes to neutrophil mobilization and host defense. In solid tumor microenvironments, sustained CXCL3 expression may promote recruitment of tumor-associated neutrophils and contribute to tumor-associated inflammation and immune regulation. Therefore, CXCL3 is an important research target for CXCR2-dependent neutrophil chemotaxis, inflammation, and tumor microenvironment studies.

CXCL3 is a soluble ELR-positive CXC chemokine that activates CXCR2 on myeloid cells and primarily promotes neutrophil chemotaxis. CXCL3-mediated leukocyte recruitment links inflammatory and tumor-associated signals with changes in local myeloid cell infiltration. It is involved in acute inflammatory responses and has also been associated with the progression of myeloid cell-rich solid tumors. Altered CXCL3 activity can influence neutrophil recruitment in inflammatory and tumor tissues. Overall, CXCL3 represents an important research target for studying ELR-positive CXC chemokine signaling and CXCR2-dependent myeloid cell recruitment.

CXCL3 Protein Function: Core Roles in Myeloid Chemotaxis and Acute Inflammatory Tuning

The biological functions of secreted ELR-motif CXCL3 chemokine are fully focused on high-affinity CXCR2 receptor binding and myeloid inflammatory signal amplification:

  • CXCR2 High-Affinity Agonist: Potently binds CXCR2 to trigger robust neutrophil migration responses.
  • Acute Infection Recruitment: Mobilizes circulating myeloid cells to sites of bacterial infection injury.
  • Tumor Myeloid Homing: Draws immunosuppressive TANs into malignant lesion microenvironments.
  • Host immune homeostasis regulation: CXCL3 is briefly induced to express when the body is slightly infected, which can quickly recruit myeloid immune cells to infiltrate the focus, effectively remove invasive pathogens and damaged tissue fragments, help inflammation subside and maintain local immune homeostasis.
  • Pathological correlation of disease: abnormal excessive secretion of CXCL3 will aggravate multiple organ injury caused by sepsis, and at the same time continue to recruit immunosuppressive neutrophils, mediate immune escape from tumor microenvironment, and promote malignant progress of solid tumor.
