Desmoglein 2 (DSG2) is a single pass transmembrane desmosomal cadherin encoded by DSG2 gene. This protein localizes to specialized plasma membrane domains which assemble into desmosome intercellular junctions within epithelial cell populations. DSG2 contains multiple extracellular cadherin type repeat domains, a transmembrane helix and a large intracellular cytoplasmic tail domain. Extracellular repeat domains project toward intercellular space, and calcium binding sites distribute across these extracellular modules. Its cytoplasmic tail faces cell interior and carries sequence motifs dedicated to binding desmosome specific intracellular adaptor molecules. Newly translated DSG2 undergoes folding and quality control inside endoplasmic reticulum compartments, and correctly processed protein is transported toward plasma membrane to concentrate at nascent desmosome assembly sites. DSG2 expression profile shows cell type specificity, and its protein abundance correlates with the density of desmosome junction structures formed between adjacent epithelial cells. Only properly folded polypeptide can accumulate at cell surface junction sites, while misprocessed material will be cleared by intracellular quality control systems.
Within desmosome junctions, DSG2 extracellular domains mediate heterophilic intermolecular contacts with desmocollin family cadherin molecules presented on opposing cell surfaces, and calcium ions preserve extracellular domain conformation required for stable molecular recognition. Its large intracellular cytoplasmic tail recruits desmosomal adaptor proteins to build dense multi protein plaque assemblies beneath plasma membrane. These cytoplasmic plaque complexes further anchor to intermediate filament cytoskeleton networks, generating robust mechanical linkage across intercellular desmosome junctions. Other cadherin superfamily members belong to adherens junction pathway and cannot substitute for desmosome specific molecular functions performed by DSG2. Variations in DSG2 surface abundance directly affect desmosome junction formation capacity. Structural alteration within extracellular cadherin repeats or cytoplasmic adaptor binding interfaces may interfere with heterophilic contact or intracellular plaque assembly and weaken overall desmosome based intercellular mechanical connectivity. Changed abundance of DSG2 also influences how readily new desmosome structures can form when epithelial cells establish new cell cell contacts.

The biological functions of DSG2 are focused on desmosomal heterophilic cell cell recognition, intracellular desmosome plaque assembly and intermediate filament anchoring:
Creative Biolabs offers purified DSG2 membrane samples via standardized preparation workflows, including full length DSG2 constructs and isolated extracellular structural variants. Isolated extracellular fragments may not support complete heterophilic adhesion and cytoplasmic plaque recruitment related behaviours, while full length constructs may be suited for desmosomal cadherin mediated junction assembly related research. All samples receive routine quality screening, and functional relevant observation may only be carried out with full length samples under simulated membrane environments. All sample batches follow unified processing standards to maintain consistent structural features for comparative laboratory analysis across separate test groups.
