Background

F-box and WD repeat domain containing 7 (FBXW7) is a cytoplasmic substrate-recognition adaptor encoded by FBXW7 gene, broadly expressed across somatic cell populations, localized to cytoplasmic and perinuclear membrane compartments. Distinct from other F-box paralogs, FBXW7 relies on conserved WD40 repeat domains to recognize phosphorylated oncogenic substrates, while simultaneously mediating substrate recruitment to SCF ubiquitin ligase complexes to trigger proteasomal degradation cascades. Under resting physiological immune states, basal FBXW7 recognition activity sustains balanced turnover of proliferative mediators without disrupting normal cell cycle progression. Upon oncogenic activation and metabolic stress, accumulated phosphorylated target substrates are captured by FBXW7 adaptor modules, facilitating ubiquitination and downstream clearance of proliferation-driving factors. Unlike broad-spectrum ubiquitin adaptors with general protein turnover roles, FBXW7 bears specialized tumor-suppressive substrate recognition functions that cannot be fully compensated by other F-box family members, linking substrate phosphorylation directly to oncogene degradation pathways. Loss of functional FBXW7 severely blunts oncoprotein clearance and accelerates aberrant cell proliferation, while sustained FBXW7 adaptor activity restrains malignant transformation, establishing FBXW7 as a core research target for ubiquitin proteolysis biology and anti-tumor compound screening.

FBXW7 executes core substrate-recruiting ubiquitin adaptor function within cytoplasmic membrane-associated complexes, utilizing conserved WD40 substrate-binding pockets to power selective capture of phosphorylated proliferative target proteins. Conserved phospho-peptide recognition motifs exclusively bind oncogenic degradation substrates, separating its substrate spectrum from housekeeping ubiquitin adaptors distributed throughout cellular compartments. FBXW7-mediated substrate shuttling bridges intracellular kinase signaling and proteasomal degradation machinery, balancing proliferative signal intensity and oncoprotein turnover efficiency according to cellular stress load. FBXW7 participates in extensive physiological processes including cell cycle checkpoint control, proliferative factor clearance and tumorigenesis suppression. Deficient FBXW7 adaptor capacity disrupts oncogenic protein surveillance and impairs intrinsic growth restriction mechanisms. Therefore, FBXW7 represents a pivotal research target for SCF complex carrier study and anti-proliferative therapeutic screening.

FBXW7 Protein Function: Core Roles in Phospho-Substrate Recognition and Tumor Suppressive Tuning

The biological functions of FBXW7 are focused on phosphorylated substrate capture, SCF ligase recruitment and cell cycle signal control:

  • Phospho-Dependent Substrate Binding: Captures phosphorylated oncogenic mediators via conserved WD40 repeat domains.
  • Ubiquitin Ligase Recruitment: Delivers bound substrates to SCF complexes to initiate proteasomal degradation.
  • Cell Cycle Homeostasis Maintenance: Sustains controlled turnover of proliferative proteins under basal physiological conditions.
  • Tumor Suppressive Modulation: Governs the magnitude of oncoprotein clearance upon cellular transformation stress.
  • Disease Relevance: FBXW7 loss drives accumulation of oncogenic substrates and tumor progression, while intact adaptor activity restrains malignant proliferation.
