FMS-like tyrosine kinase 3 (FLT3, also known as CD135) is a receptor-type tyrosine kinase characterized by an extracellular ligand-binding region, a transmembrane domain, and an intracellular tyrosine kinase domain. It is primarily expressed in early hematopoietic stem and progenitor cells, particularly those committed to dendritic cell (DC) development. Upon binding its cognate ligand, FLT3LG, FLT3 undergoes receptor dimerization, which activates its intracellular kinase domain and initiates downstream signal transduction.
Unlike many broadly distributed hematopoietic receptors, FLT3 signaling plays a highly specific and irreplaceable role in regulating the proliferation, differentiation, and survival of DC progenitors. Under steady-state conditions, baseline FLT3 activity is essential for maintaining normal hematopoietic homeostasis and DC populations. Fluctuations in extracellular FLT3LG levels directly modulate FLT3 activation, thereby dictating the expansion efficiency and subset distribution of DCs. Because its functional disruption profoundly alters antigen-presenting cell dynamics, FLT3 remains a critical focal point for research into hematopoietic receptor signaling and early immune cell ontogeny.
