HSPA1B (a member of heat shock protein family A(Hsp70)1B) is a cytoplasmic soluble molecular chaperone encoded by HSPA1B gene, which is widely expressed in the cytoplasmic water-soluble region of somatic cells. There are essential structural differences between the protein and the integrated transmembrane scaffold protein, and its amino acid sequence does not contain lipid-anchored transmembrane domains and membrane-binding functional motifs. It is a typical cytoplasmic soluble molecular chaperone, which can bind unfolded polypeptide substrates under physiological steady-state conditions and participate in the folding regulation of cell basic proteins. When the function of intracellular molecular chaperone folding auxiliary pathway is insufficient, improperly folded peptides are prone to aggregation and deposition, while HSPA1B can maintain the dynamic balance of polypeptide metabolism turnover in different microenvironments through mild protein folding buffer effect. Due to the different kinds of cytoplasmic partitions of cells will produce different misfolded polypeptide substrates, enough soluble molecular chaperones are needed in the cells to ensure the steady state of the whole protein folding system; HSPA1B dispersed in cytoplasm can continuously bind to the hydrophobic fragments exposed by peptides, effectively inhibit abnormal protein aggregation and maintain protein homeostasis of cells.
Sequence variation of the HSPA1B gene may affect substrate binding or chaperone activity and thereby influence cellular protein folding. HSPA1B encodes a stress-inducible Hsp70 molecular chaperone that is highly homologous to HSPA1A and shares a conserved ATP-dependent mechanism of substrate binding and release. Other cytoplasmic Hsp70 family members, including HSPA8, also participate in protein folding, refolding, and proteostasis. HSPA1B expression increases in response to proteotoxic and other cellular stresses, making it a useful research target for studying stress-inducible molecular chaperone function and intracellular protein quality control. HSPA1B is a soluble intracellular protein that is predominantly cytoplasmic under basal conditions but can redistribute between cellular compartments in response to stress. Its binding to unfolded or misfolded polypeptides does not involve transmembrane signaling, distinguishing it from integral membrane proteins. Through ATP-dependent substrate binding and release, HSPA1B contributes to protein folding, refolding, and the prevention of aberrant protein aggregation. Reduced HSPA1B function may impair cellular proteostasis and increase the accumulation of misfolded proteins, supporting its value in basic research on heat shock proteins and molecular chaperone biology.
Fig. 1 Schematic representation of human stress‑inducible cytosolic Hsp70 domain architecture (HSPA1A, high sequence homology to HSPA1B). The nucleotide‑binding domain and substrate‑binding domain are connected via flexible linker; substrate‑binding domain captures exposed hydrophobic segments from unfolded polypeptides to sustain cytoplasmic proteostasis balance.1
The biological functions of soluble cytosolic HSPA1B chaperone are focused on sustained unfolded polypeptide capture and intracellular protein folding coordination:
Creative Biolabs offers purified HSPA1B protein samples manufactured under unified preparation workflows, including full-length HSPA1B constructs and isolated substrate-binding domain variants. Truncated binding fragments cannot sustain complete polypeptide refolding interaction behaviors, while full-length constructs match research focused on chaperone-polypeptide association and cytoplasmic proteostasis observation. All sample batches receive unified quality screening procedures. Functional relevant observation may only proceed using full-length protein under simulated cytoplasmic microenvironment setups. Consistent structural characteristics are preserved across all batches to support comparative laboratory analysis between separate experimental groups. Full-length HSPA1B samples maintain intact substrate binding pocket conformation after standardized purification workflows, which supports reliable detection of weak and transient binding events between the chaperone and assorted unfolded polypeptides in comparative functional analysis.
