Interleukin 15 receptor subunit alpha (IL15RA) is a single pass transmembrane cytokine receptor subunit encoded by IL15RA gene. This protein localizes to plasma membrane surface of multiple immune cell populations. IL15RA contains large extracellular cytokine binding domain, one transmembrane helix and a short intracellular cytoplasmic segment. Extracellular domain is exposed to extracellular milieu while cytoplasmic segment faces cell interior. Alternative transcript isoforms are generated, including secreted isoform variants lacking transmembrane region. Polypeptide translation and folding take place within secretory organelles, and properly folded full length isoform traffics toward plasma membrane. Different immune cell subtypes show variable IL15RA expression magnitude. Only correctly folded extracellular domain can accomplish high affinity cytokine ligand engagement, while mis folded polypeptide will be cleared by intracellular quality control machineries. Secreted isoform generation is controlled by transcript processing and can change total pool of soluble receptor molecules in extracellular compartments.
The extracellular domain of membrane bound IL15RA establishes high affinity physical contact with interleukin 15 cytokine molecules. This receptor subunit can present bound cytokine ligand toward other receptor subunits to assemble complete multi subunit cytokine receptor complexes. Short intracellular cytoplasmic segment possesses limited intrinsic signal triggering capacity on its own, and full downstream signal output depends on assembly with additional partnering receptor subunits. Secreted isoform variants can also bind soluble cytokine molecules inside extracellular compartments. Other cytokine receptor subunits cannot reproduce the high affinity interleukin 15 binding property provided by IL15RA. Variation in IL15RA abundance changes cellular capacity to capture and present corresponding cytokine ligand molecules. Sequence modification within extracellular ligand binding domain may reduce or abolish interleukin 15 binding affinity, while alteration of cytoplasmic segment sequence influences intracellular molecular interactions after multi subunit complex formation. Shifts in the ratio between membrane bound and secreted isoform can modify overall cytokine sequestration behaviour in local extracellular environments.

The biological functions of IL15RA are focused on high affinity interleukin 15 ligand capture, cytokine ligand presentation and multi subunit cytokine receptor complex assembly:
Creative Biolabs offers purified IL15RA membrane samples via standardized preparation workflows, including full length IL15RA constructs and isolated extracellular ligand binding domain variants. Isolated extracellular fragments may not support complete ligand presentation and multi subunit complex assembly related behaviours, while full length constructs may be suited for cytokine receptor subunit mediated ligand capture associated research. All samples receive routine quality screening, and functional relevant observation may only be carried out with full length samples under simulated membrane environments. All sample batches follow unified processing standards to maintain consistent structural features for comparative laboratory analysis across separate test groups.
