Interleukin 18 binding protein (IL18BP) is a secreted soluble polypeptide encoded by IL-18 BP gene, which can be secreted by various stromal cells and immune cells and released into the extracellular space. The full-length sequence of the protein does not contain hydrophobic transmembrane helix and membrane anchoring structural domain, and has no cell membrane localization ability. It can be used as a high-affinity extracellular bait ligand, specifically binding mature IL-18 cytokines, and is a key antagonist molecule for regulating IL-18 immune signals. A large number of studies on the binding mechanism have confirmed that IL-18 BP and IL-18 can form a stable complex through electrostatic interaction and hydrophobic interaction, effectively blocking the specific binding of IL-18 with functional receptors on the cell membrane surface, thus blocking the initiation of downstream immune signals.
In the absence of IL-18 BP antagonistic regulation, IL-18-mediated signaling pathway will continue to be over-activated, causing abnormal amplification of immune cascade. By limiting the bioavailability of extracellular IL-18, IL-18 BP inhibits the excessive inflammatory activation effect, and maintains the inflammatory homeostasis and immune basic balance of mucosal and lymphoid microenvironment. The highly conserved ligand binding domain in the central region of IL-18 BP can form a wide molecular contact surface with IL-18, which can stably bind the target molecule under the condition of physiological extracellular concentration, ensuring the high efficiency and reliability of bait antagonistic effect, and has important physiological significance for maintaining immune homeostasis and avoiding abnormal activation of inflammation.
Sequence variants within IL18BP coding regions might weaken IL-18 binding affinity and disrupt negative immune feedback loops observed in primary tissue models. No other soluble decoy protein replicates the high ligand affinity of IL18BP for IL-18, though other IL-1 family binding factors carry partial overlapping inhibitory capacity. IL18BP exists exclusively as secreted extracellular protein without membrane integration or cytoplasmic retention. Its fully soluble secreted localization grants dual roles in sequestering free IL-18 and restricting receptor-mediated immune signal transduction. Reduced IL18BP secretion elevates unbound extracellular IL-18 concentrations and amplifies inflammatory cascade activity, rendering this decoy polypeptide a suitable research target for cytokine immune homeostasis analysis.
