Background

Integrin subunit alpha 2 (ITGA2) is a single pass transmembrane integrin alpha subunit encoded by ITGA2 gene. This protein localizes to plasma membrane of mesenchymal, epithelial and migratory cell populations within connective tissues. ITGA2 contains large extracellular ligand binding domains, a transmembrane helix, and a short intracellular cytoplasmic segment. ITGA2 cannot reach cell surface as a mature functional molecule without forming heterodimers with compatible beta integrin subunits. Heterodimer assembly takes place inside intracellular compartments before protein trafficking to plasma membrane. Unpaired ITGA2 subunits are retained intracellularly and are not efficiently transported to cell surface.

Once assembled into heterodimer complexes at plasma membrane, ITGA2 containing integrins establish non‑covalent molecular contacts with defined components of extracellular matrix. The intracellular cytoplasmic segment of the heterodimer provides docking sites for cytoplasmic partner proteins after extracellular molecular binding occurs. Different integrin alpha subunits possess distinct ligand binding preferences. ITGA2 containing heterodimers recognize a specific subset of extracellular matrix molecules that are not fully recognized by other integrin alpha paralogs. Variations in ITGA2 expression level may change the abundance of corresponding heterodimers at cell surface and alter the capacity for cell‑matrix molecular recognition.

ITGA2 Protein Function: Core Roles in Extracellular Matrix Recognition and Membrane Heterodimer Assembly

The biological functions of ITGA2 are focused on extracellular matrix molecule recognition, integrin heterodimer formation and cell matrix interface modulation:

  • Matrix oriented Binding Surfaces: The extracellular globular domain might mediate non‑covalent contacts with subsets of extracellular matrix molecular components upon formation of heterodimer complexes.
  • Heterodimer Assembly Partner: Requires pairing with compatible beta integrin subunits to build stable membrane anchored integrin assemblies.
  • Cell Matrix Interface Modulation: Could support cell surface anchoring toward surrounding matrix components to shape cell attachment status.
  • Membrane Complex Scaffolding: The intracellular region is proposed to recruit cytoplasmic molecular partners after stable extracellular molecular engagement.
  • Research Model Relevance: Structural alteration of ITGA2 may modify heterodimer assembly and matrix recognition behaviours within laboratory research systems.

ITGA2 Membrane Protein Product

Creative Biolabs offers purified ITGA2 membrane samples via standardized preparation workflows, including full‑length ITGA2 constructs and isolated extracellular structural variants. Isolated extracellular fragments may not support complete heterodimer assembly and matrix binding related behaviours, while full‑length constructs may be suited for integrin subunit pairing and extracellular matrix molecular interaction related research. All samples receive routine quality screening, and functional relevant observation may only be carried out with full‑length samples under simulated membrane environments. All sample batches follow unified processing standards to maintain consistent structural features for comparative laboratory analysis across separate test groups.
