Background

Integrin subunit alpha X (ITGAX, also known as CD11c) is a transmembrane integrin α subunit encoded by the ITGAX gene. ITGAX preferentially assembles with ITGB2 (CD18) to form the heterodimeric integrin αXβ2 (CR4). This receptor is prominently detected on myeloid populations including dendritic cells, macrophages and monocytes, with low expression also observed in certain lymphocyte subsets.At the cell surface, αXβ2 mediates cell adhesion and migratory behaviors via binding extracellular ligands such as fibrinogen and ICAM family molecules. Under steady-state physiological conditions, basal ITGAX expression supports low-level leukocyte anchoring. Upon inflammatory stimulation and pathogen exposure, ligand-bound αXβ2 initiates intracellular signaling cascades to facilitate myeloid cell recruitment, substrate uptake and downstream antigen processing.

As a member of the β2 integrin subfamily, αXβ2 mediates myeloid cell trafficking, opsonized substrate recognition and antigen-presenting cell maturation. Altered ITGAX expression disrupts leukocyte adhesion and tissue infiltration, triggering abnormal inflammatory responses. Embedded in plasma membranes, αXβ2 uses conserved extracellular domains to bind ligands and activate intracellular cytoskeletal signaling, adjusting migratory activity based on inflammatory stimuli. Loss of ITGAX impairs systemic innate immune surveillance, making it an important research target for integrin biology and inflammatory cell migration studies.
