Background

Jagged canonical Notch ligand 1 (JAG1) is a single-pass transmembrane ligand encoded by the JAG1 gene, widely expressed in epithelial and mesenchymal progenitor cells, localized exclusively to plasma membrane lipid bilayers. Distinct from Delta-type Notch ligands, JAG1 extracellular EGF-repeat domains bind Notch receptor ectodomains to trigger intracellular NICD cleavage and stem cell fate transcriptional cascades. Under steady developmental conditions, basal JAG1 expression maintains balanced progenitor cell renewal without premature terminal differentiation. Upon tissue morphogenesis signals, cell-surface JAG1-receptor ligation releases active NICD fragments to drive lineage specification and tissue boundary formation programs. Unlike transient secreted signaling factors, JAG1 bears contact-dependent developmental regulatory functions that cannot be fully compensated by other Notch ligands, linking cell-cell contact directly to progenitor differentiation pathways. Loss of functional JAG1 disrupts tissue patterning and impairs stem cell pool maintenance, while sustained JAG1 signaling sustains undifferentiated progenitor states, establishing JAG1 as a core research target for Notch ligand biology and developmental modulator screening.

JAG1 executes membrane-bound Notch ligand function embedded within cell surface bilayers, utilizing tandem EGF-like extracellular motifs to form stable trans-interactions with Notch receptor ectodomains. Conserved ligand-binding arrays exclusively recognize canonical Notch receptors, separating its binding spectrum from unrelated cell-surface signaling ligands. JAG1-mediated juxtacrine signaling bridges intercellular contact and nuclear NICD transcriptional programming, balancing progenitor self-renewal and differentiation according to tissue developmental stage. JAG1 participates in embryonic tissue patterning, adult stem cell maintenance and vascular cell lineage control. Deficient JAG1 expression ablates canonical Notch signal transduction and disturbs organ morphogenesis. Therefore, JAG1 represents a pivotal research target for juxtacrine ligand study and stem cell compound screening.

JAG1 Protein Function: Core Roles in Notch Receptor Binding and Progenitor Fate Tuning

The biological functions of JAG1 are focused on Notch ectodomain recognition, juxtacrine signal initiation and stem cell control:

  • Notch Receptor Binding: Interacts with cell-surface Notch receptors via extracellular EGF repeat arrays.
  • NICD Cleavage Triggering: Induces receptor proteolysis to release intracellular active Notch fragments.
  • Tissue Homeostasis Maintenance: Sustains balanced progenitor renewal under resting tissue status.
  • Lineage Differentiation Modulation: Adjusts stem cell fate commitment during morphogenesis.
  • Disease Relevance: JAG1 deficiency disrupts organ development, while overexpression locks cells in undifferentiated progenitor phenotypes.
