The LAG3 gene is located in the region of human chromosome 12p13.32 and encodes the lymphocyte activation gene 3 protein (LAG-3), which belongs to the immunoglobulin superfamily. LAG-3 is a transmembrane glycoprotein composed of an extracellular region, a transmembrane region, and an intracellular region. It is mainly expressed on the surfaces of immune cells such as activated T cells, natural killer cells, and dendritic cells. As an important immune checkpoint molecule, LAG-3 negatively regulates the proliferation, activation, and cytokine secretion of T cells by binding to its ligands (such as MHC-II molecules). It plays a crucial role in maintaining immune homeostasis and preventing autoimmune reactions. This gene has multiple single nucleotide polymorphism loci. Some functional variations can affect the protein expression level or ligand binding affinity, thereby altering the intensity of immune inhibitory signals. Moreover, changes in LAG-3 expression levels are closely related to tumor immune escape, chronic viral infections, and the occurrence and development of autoimmune diseases.
